Giuseppe Brotzu

Giuseppe Brotzu (1895–1976) was an Italian pharmacologist and politician who discovered the cephalosporin class of antibiotics, a cornerstone of modern medicine. His work in the 1940s on a fungus from seawater off Sardinia led to the isolation of cephalosporin C, which later gave rise to broad-spectrum antibiotics.

Early life and career

Giuseppe Brotzu was born in Ghilarza, Sardinia, in 1895. He studied medicine at the University of Cagliari and later specialized in hygiene and public health. In the 1930s, he became a professor of hygiene at the University of Cagliari and also served as the director of the city's public health laboratory. His early work focused on sanitation and infectious diseases, which led him to investigate natural sources of antibacterial substances.1

Discovery of cephalosporin

In 1945, Brotzu noticed that sewage outfalls along the Sardinian coast were free of typhoid bacilli, despite heavy contamination. He hypothesized that a microorganism in the seawater produced an antibacterial compound. He isolated a fungus, later identified as Cephalosporium acremonium, from the water and found that its culture filtrate inhibited the growth of Salmonella typhi and other bacteria. He published his findings in a local journal in 1948, but the work was largely ignored internationally.2

Collaboration with Oxford and development of cephalosporins

Brotzu sent samples of the fungus to the Sir William Dunn School of Pathology at Oxford University, where Howard Florey and his team, including Edward Abraham, took up the research. They isolated cephalosporin C, which had a beta-lactam ring and was resistant to penicillinase. This led to the development of the cephalosporin class of antibiotics, which are now among the most widely used antibiotics worldwide. Brotzu's contribution is recognized as a pivotal step in antibiotic history.3

Political and public health roles

Brotzu was also active in politics. He served as mayor of Cagliari from 1944 to 1946 and later as a member of the Italian Parliament. He was a member of the Italian Socialist Party and later the Italian Democratic Socialist Party. His political career was intertwined with his public health work, as he advocated for better sanitation and healthcare in Sardinia.4

Lesser-known aspects

Brotzu's discovery was initially published in an obscure Sardinian journal, and it was only through his persistence that the samples reached Oxford. He also isolated other antibiotic-producing organisms from the same environment. Brotzu's work was not fully recognized during his lifetime, but he received the honorary degree from the University of Oxford in 1971. The fungus he discovered was later reclassified as Acremonium chrysogenum. Brotzu's legacy is also preserved in the Brotzu Hospital in Cagliari, named in his honor.5
