Charles-Édouard Brown-Séquard (1817–1894) was a Mauritian-born physiologist and neurologist whose experiments reshaped understanding of the nervous system and endocrine glands. He is best known for describing the Brown-Séquard syndrome, a set of motor and sensory deficits resulting from a spinal cord hemisection, and for his controversial self-injection of testicular extracts, which helped launch the field of organotherapy and, indirectly, modern endocrinology.
Brown-Séquard was born in Port Louis, Mauritius, to an American father and a French mother. He studied medicine in Paris, earning his doctorate in 1846. His early research on the spinal cord and reflex actions established his reputation, and he later held positions in the United States, Britain, and France. He was a founding editor of the Journal de la physiologie de l'homme et des animaux and served as a professor at the Medical College of Virginia and Harvard University before returning to Paris.
In 1851, Brown-Séquard described the clinical and experimental consequences of a hemisection of the spinal cord. He demonstrated that damage to one half of the cord causes ipsilateral paralysis and loss of proprioception, with contralateral loss of pain and temperature sensation. This dissociation of sensory modalities provided crucial evidence for the crossing of sensory pathways in the spinal cord. The syndrome remains a classic neurological teaching point and is still used to localize spinal cord lesions.
In 1889, at age 72, Brown-Séquard reported to the Société de Biologie in Paris that he had injected himself with extracts of dog and guinea pig testes, claiming increased vigor and mental acuity. Although the effects were likely placebo, his report ignited widespread interest in organotherapy and stimulated research into internal secretions. This work, though controversial, is often cited as a precursor to hormone replacement therapy and the discovery of testosterone. He also studied the adrenal glands, suggesting they were essential for life, and conducted experiments on the thyroid.
Beyond his famous syndrome, Brown-Séquard made several other contributions. He was one of the first to describe the vasomotor effects of the sympathetic nervous system and conducted pioneering research on epilepsy, showing that certain brain lesions could induce seizures. He also studied the physiology of the skin and the effects of temperature on nerve conduction. His career was marked by frequent relocations between continents, and he was known for his intense, sometimes combative, personality. He died in Paris in 1894, leaving a legacy that bridges neurology and endocrinology.
Brown-Séquard's work, though sometimes dismissed in his time, laid foundations for both neurology and endocrinology.
